We developed an assay (MOCA) for compounds binding to G protein coupled receptors (GPCRs) or transporters. MOCA involves translocation of a GPCR or transporter into which we incorporate a nuclear localization sequence (NLS), from the cell surface. Binding of compounds to the GPCR-NLS or transporter-NLS prevents the translocation. The method does not depend on any knowledge of G protein.
